I'm pretty sure that the mameui32.ini contains settings for the Mame 32 Frontend. If you look at the contents you could verify it. Most people use Mame from mamedev.org. There is not much point in running a frontend version of Mame within GameEX which of course is a frontend itself.

You can also use and advanced config to watch for additional processes.

To be clear, if the emulator has command line support then getting it going should be 'a snap'. If not it would require that a loader be written. Edit: It should work from the command line.
